   目的:为了探讨和研究不同产地茯苓和葛根中19种氨基酸的含量和差异。方法:将样品加入6mol/l盐酸10ml于50ml玻璃试管于150℃鼓风干燥机水解1小时。加入0.1mol/l盐酸稀释后定容至10ml试管。采用氨基酸分析仪法,以茚三酮为柱后衍生化试剂,以Na离子分离柱进行梯度洗脱。结果:不同产地茯苓和葛根中所含氨基酸,包括牛磺酸(Tau)、天门冬氨酸(asp)、苏氨酸(Thr)、丝氨酸(Ser)、谷氨酸(Glu)、脯氨酸(Pro)、甘氨酸(Gly)、丙氨酸(Ala)、缬氨酸(Val)、甲硫氨酸(Met)、异亮氨酸(Tle)、亮氨酸(Leu)、酪氨酸(Try)、苯丙氨酸(Phe)、组氨酸(His)、色氨酸(Trp)、赖氨酸(Lys)、精氨酸(Arg)、胱氨酸(Cys)的浓度在9.980~168ug/ml范围内呈现良好线性关系(r =0.9930~0.9999);平均回收率在 80~100% 之间。结论:不同产地的茯苓和葛根所含氨基酸种类、数量不同。分别对茯苓、葛根不同产地同一氨基酸,同一氨基酸不同产地进行方差分析,根据数据显示其具有统计学意义。对不同产地茯苓、葛根同一氨基酸进行聚类分析,可直观了解茯苓、葛根中不同氨基酸含量差异。

The content difference of 19 kinds of amino acids in poria cocos and pueraria
Abstract
Objective:In order to study the difference of 19 kinds of amino acids in poria and pueraria.Methods:To add samples to 6 mol/l HCL 10 ml in 50ml glass tube in 150 ℃ blast drier hydrolysis for 1 hour,And add 0.1mol/l hydrochloric acid and dilute it to 10ml test tube.An amino acid analyzer was used to determine the gradient elution of the column with the Na ion separation column.Result:Poria cocos and puerarin amino acid contained in different regions, including the taurine (Tau), aspartic acid (asp), threonine (Thr), serine (Ser) and glutamate (Glu), proline (Pro), glycine (Gly), alanine (Ala), valine (Val), methionine (Met), isoleucine (Tle), leucine (Leu), tyrosine (Try), phenylalanine (Phe) and histidine (His), tryptophan (Trp), lysine (Lys), arginine (Arg), cystine (Cys) concentration within the scope of 9.980 to 168 ug/ml show good linear relationship (r = 0.9930 ~ 0.9999);The average recovery is between 80 and 100%.Conclusion:There are different kinds of amino acids and the number of amino acids.The analysis of the variance of the same amino acid and the same amino acid in different producing areas of poria cocos and kudzu was statistically significant according to the data.The different amino acid content of poria cocos and pueraria root were analyzed by clustering analysis.
