内容摘要:研究大孔树脂精制PCB醇提液中总黄酮,水提液中多酚、多糖的工艺条件。方法:把PCB的总黄酮吸附率和解析率作为指标,研究4种型号完全不同的大孔树脂对PCB总黄酮的吸附力及与解析力,确认D101作为最适树脂。优化D101大孔树脂的纯化条件;将PCB多酚、多糖吸附率与解析率作为指标,研究5种不同类型大孔树脂对PCB多酚、多糖的吸附力和解析力,将AB-8作为最适树脂。优化AB-8大孔树脂纯化条件。结果:PCB总黄酮分离富集条件为:上样速度2 BV/h,上样量120.00 mL,上样浓度为4.890 mg/mL,吸附1.5 h,除杂溶剂4%乙醇,除杂速度3 BV/h,用量256.41 mL,将60%乙醇作为洗脱溶剂,3 BV/h的洗脱速度,125.00 mL洗脱剂用量,经D101 处理后PCB总黄酮纯度至55. 25% ;多酚、多糖的分离富集条件为:上样速度2 BV/h,上样量15.00 mL,上样浓度为0.5 g/mL(生药/溶剂),1.5 h吸附,40%乙醇作为洗脱溶剂,3 BV/h的洗脱速度,90.00 mL洗脱剂用量,经AB-8处理后的PCB多酚、多糖纯度为10.76%、40.92%,结论:D101型的大孔吸附树脂能较好将PCB总黄酮纯化富集;AB-8型大孔树脂能较好将PCB多酚、多糖纯化富集。

Study on purification technology and in vitro efficacy of PCB Jiangtang tablets
Abstract: the research of macroresin PCB ethanol extract of total flavonoids in liquid water extraction of polyphenols, process conditions of polysaccharides. Methods: the total flavone adsorption rate and resolution rate of PCB as index, adsorption capacity of macroporous resin on the 4 types of different PCB total flavonoids and resolution D101, recognized as the optimum resin. Optimum purification conditions of D101 macroporous resin; PCB polyphenols, rate and desorption rate as the index of adsorption of polysaccharide, 5 different types of macroporous resin on PCB adsorption capacity and resolution of polyphenols, polysaccharides, AB-8 as the optimum resin. The optimized AB-8 Kong Shuzhi purification conditionResults: PCB flavonoids separation and enrichment conditions were the sample rate of 2 BV/h, sample volume of 120 mL, the concentration of sample was 4.890 mg/mL, 1.5 h of adsorption, impurity removal rate of solvent was 4% ethanol, 3 BV/h, 256.41 mL dosage, 60% ethanol as elution solvent, elution rate of 3 BV/h, 125 mL eluting agent dosage, after treatment with D101 PCB to 55.25% purity of total flavonoids; polyphenol, polysaccharide separation and enrichment conditions were the sample rate of 2 BV/h, sample volume 15ML, the concentration of sample was 0.5 g/mL (crude drug / solvent), 1.5 h adsorption, 40% ethanol as elution solvent, elution rate of 3 BV/h, 90 mL, eluent dosage, polyphenol by PCB after AB-8 treatment, the purity of polysaccharide was 10.76%, 40.92%, conclusion: D101 macroporous resin can be used to the purification of total flavonoids from PCB enrichment; AB-8 macroporous resin could be PCB polyphenol, polysaccharide separation and purification.
